Ion Metallic Affinity Chromatography and Purification of Bacterial Toxin

چکیده
The history of the scientific development of the Chromatography as a chemical separation tool cannot be considered ancient, as the proper establishment of Chemistry as a science is considered modern when compared to the other sciences developed before it. The first literary description of the Chromatography separation technique development is attributed to the works or Mikhail Tswet in 1890 by some authors, where the chemical separation of plant extracts was conducted inside laboratory and first established, permitting the chemical separation of plant chemical substances according to their chemical characteristics of polarity and chemical affinity to the stationary phase. At that time, due to the color attributed to the plant extracts separated experimentally, the technique was named as “chromatography”, where chromato means color and graphy means registering or writing. Although the many chemical substances and bio molecules that can be separated by this technique have no color, the name was kept and the technique recognized as one of the most efficient and applicable for laboratory purification and analytical processes.

In this study Escherichia coli DE3 containing expression vector (pET21a) with cloned Persian sturgeon growth hormone (psGH) gene was grown in 10 mL LB broth on a 150 rpm shaker, at the temperature of 37 °C. At the late log phase (determined by OD standard curve) 100 &muL isopropyl &beta-D-1-thiogalactopyranoside (IPTG) was added for induction of GH synthesis. Samples were taken every 2 hours an...
متن کاملAlpha Toxin Purification and Antibody Production Against Local Strain of Clostridium septicum NH2
BACKGROUND: Clostridium septicum has played a significant role as a causative agent of many acute fetal diseases in man and animals. Alpha- toxin is the main factor in the pathogenesis of C. septicum with hemolytic, necrotic and lethal activities. OBJECTIVES: The study was designed to evaluate alpha-toxin purification and antibody production rate against a local strain of C. septicum NH2 which ...

متن کاملExpression, purification, and immunization of a chimeric protein containing immunogenic regions of flagellin and intimin proteins against E. coli O157: H7
Introduction: Enterohemorrhagic Escherichia coli (EHEC) and serotype O157: H7 is one of the most important diseases causing diarrhea. Shiga-like toxin secreted by the bacteria destroys epithelial cells and, in acute cases, causes hemolytic uremic syndrome (HUS). Antibiotic therapy is not effective against this pathogen, because it increases the production of Shiga toxin. Designing chimeric immu...
